Com a Proclamação da República em 15 de novembro de 1889, é instituído um governo republicano que, entre outras mudanças, promulga a Constituição de 1891. A partir dela, a Câmara dos Senadores, órgão vitalício do Poder Legislativo imperial, é substituída pelo Senado Federal, que, juntamente com a Câmara dos Deputados, passa a compor o Congresso Nacional, instituição em que seus integrantes representam o povo brasileiro e se elegem, por meio do voto.
O Senado Federal, todavia, não permanece o mesmo desde a sua instituição, dadas as mudanças constitucionais posteriores e as dissoluções resultantes de períodos de exceção que caracterizaram a história brasileira. Apesar dos desafios enfrentados, com a redemocratização em 1985 e a promulgação da Constituição Federal de 1988, a instituição consolida sua posição de representante dos Estados e do Distrito Federal, ao legislar, revisar, controlar e fiscalizar as leis nacionais, além de ser responsável também pela aprovação de membros que irão compor diferentes funções no Estado Brasileiro. Passa a exercer, assim, um papel amplo e atuante dentro do Poder Legislativo e da democracia brasileira.
